Laboratory Notebook: Mail from Cal [Calvin Harvey], 1989 - 1992

 File — Box: 06, Folder: 01-02
Identifier: CWG-01-022
Scope and Contents Much of the notebook is an email and letter exchange between Harley and Greider. Begins with good news, as the ribosomel RNA probe that she used seemed to have worked on the blots but not the gels. There is some bad news however, such as shattered tubes, and samples of DNA that have been used up. Grieder gave suggestions on how to run the remaining samples. Harley’s response was that Harley was analyzing telomere activity in the brain, specifically those with alzhiemers. The deeper the...

Laboratory Notebook: Yeast, Linear, Plasmids, 1988 - 1989

 File — Box: 06, Folder: 03
Identifier: CWG-01-024
Scope and Contents The notebook begins with Greider doing a large scale digest of each plasmid. The E. coli strain that Greider was using was enclosed inside the plasmid and included with it a detailed restriction enzyme map. Grieder then clipped a vector from the linear plasmid and ran it through a gel, which resulted in degraded DNA.Because of the degraded DNA Greider decided to use cells from a colleague. She harvested them and added DMSO, shich inhibits reactions in the DNA during PCR,and...

Laboratory Notebook: TLE, Early Notebooks/Telomere Cloning, 1984 - 1992

 File — Box: 06, Folder: 04
Identifier: CWG-01-025
Scope and Contents The notebook begins with an interesting note: Greider was taking classes in Biochemistry in 1984. Proceeded right back into experiments. Diluting and spinning cells into pellets and running minipreps. Also is spheroplasting cells, meaning Greider was destroying the outside cell wall with enzyme zymolyase. Greider spun down the nucleate and ran RNAse with nucleate through a gel. Running more gel studies, she found two bands. They seem to be upper and lower plasmid bands. Greider wanted to cut...
