Scope and Contents
The notebook contains films created from the Blackburn laboratory with x-rays inside the notebook. In IV 14, the invitro experiment was exposed for 2 days and in IV 16 the gel was exposed for 6 days. Both of which created dark columns. In IV 17, the 6 % Acryl gel was conducted and another gel in the image was done with Urea. The next gel in the notebook were both a titration and a gel. Both were exposed for five days with a ddNTP treatment. In IV 22, the three day exposure that created two large dark columns. In a oligonucleotide gel study, both IV 24 and IV 25, were exposed for thirty-six hours for each IV experiment. In another IV 25 experiment Grieder used a G4T2 DNA sequence for all columns but with different restriction enzyme. Specifcally, she used Hind, RNase, Protienase. In an interesting note, it was more important for Grieder to emphasize the lightest bands even if others are overexposed and created darker bands and columns. IV 27 was a RNA gel study that used an 8% Acryl under twenty-four hour exposure. In another RNA gel study, IV 30, Grieder exposed two gels. One was under three and a half hours and another for nine hours. In IV 32, this could be considered the longest gel study yet, as it was 60 hours and was exposed for over six days. In IV 35, the light bands in columns were exposed over twenty days with no screen. The noteook ends with another gel study but this was one is interesting as it does not use Yeast of Tetrahymena cells but used a Trypanosoma species.
