Laboratory Notebook 9: In Vitro 124-150, 1989 - 1990
Scope and Contents
The notebook begins with a graph of the counts per minute of T2G4 oligonucleotide and the same nucleotide combined with pBR enzyme. Greider then ran a program on the corresponding gel. Like previous notebooks she reviewed and remarked on invitro experiments (T2G4). Also sets up two kinds of experiments: One is a processivity experiment: mix enzyme with T2G4 and combined (T2G4 + buffer). The second is a Competition experiment: mixes enzyme with T2G4 and combined (T2G4 + ddT). Greider had a group meeting which began by anlyzing the different types of experiments and concluded that experiments in kinetics seemed to suggest that unusual enzymes interactions. Greider graphed the experiments and decides to do more processivity experiments with more sequence counts. The notebook contained several intersting documents. One of them was a series of calculations on the directions to a location, seems to be a hospital. The other is letter. Greider sent a letter to Charles McMaster. In it Greider wrote about the materials and methods section of her paper. Greider also had questions, involving calculations. McMaster eventually answers the letter, but mostly answers her questions.
